APLIKASI PCR-RAPD DALAM IDENTIFIKASI FMA

Abstract


Identifikasi FMA berdasarkan karakteristik morfologi memiliki kelemahan, yaitu tidak dapat mengungkap keragaman pada tingkat strain dalam suatu spesies, sehingga perlu memanfaatkan karakter molekular dan genetik berdasarkan DNA ribosom. Teknologi yang digunakan adalah Random Ampified Polymorphic DNA (RAPD) yang didasarkan pada metode Polimerase Chain Reaction (PCR) dengan menggunakan primer rantai pendek yang dapat diterapkan pada mikroba yang tidak dapat dikulturkan seperti FMA, melalui amplifikasi genom dari spora tunggal, akar terinfeksi, atau sampel tanah langsung dari lapangan.Teknik PCR bisa berupa Nested PCR yang untuk memonitor spesies FMA dan kelimpahannya atau Competitive PCR untuk mendeteksi sekuen yang muncul dalam FMA pada akar yang dikumpulkan dari lapangan. Dengan teknik PCR genom DNA tersedia dalam jumlah memadai untuk keperluan identifikasi.
